John Allerton apparently resided in Holland. Unlike the other Pilgrims, he planned on returning with the Mayflower to help others of the Leyden congregation make their way to America. However, his death the first winter ended those plans.
A John Allerton buried a child in St. Peters, on 21 May 1616 in Leyden, Holland. There was a Robert Allerton from Scotland that had lived in Leyden since 1607. (The England and Holland of the Pilgrims, by Henry Martyn Dexter, et al., 1905)
There is no known relation between Isaac Allerton and John Allerton, but one may very well exist since the ancestry of both is unknown. Both Isaac Allerton and John Allerton buried children in St. Peter's, Leyden, and both apparently lived in the same neighborhood. Isaac Allerton had a sister Sarah in Leyden, who married Degory Priest in 1611.
